杏丁注射液对血管内皮细胞HSP 72表达的影响

    [摘要]  目的:观察杏丁注射液对紫外线照射后血管内皮细胞热休克蛋白72(heat shock protein 72, HSP 72)表达的影响。方法:采用体外培养的猪主动脉血管内皮细胞,以含浓度分别为0.1、0.5、1.0、5.0、10 mg/ml的杏丁注射液的培养基培养72 h后,用紫外线分别照射细胞30 min,以Westernblot方法检测内皮细胞中的HSP 72表达。结果:正常情况下检测不到HSP 72表达,紫外线照射后血管内皮细胞HSP 72有较强表达,杏丁注射液在0.1 mg/ml浓度即有很明显的促进HSP 72表达的作用,1.0 mg/ml浓度时其作用达到高峰,再增加浓度并不能增加HSP 72表达。结论:杏丁注射液能通过促进HSP 72表达而保护血管内皮细胞在应激状态下免受损伤,这可能是其防治心脑血管疾病的重要机制之一。

  Effect of Xingding Injection on expression of heat shock protein 72 in vascular endothelial cells
  ABSTRACT  Objective: To study the effect of Xingding Injection on the expression of heat shock protein 72 (HSP 72) in vascular endothelial cells after ultraviolet radiation. Methods: Porcine aortic endothelial cells were cultured for 72 hours in culture mediums with different concentrations (0.1, 0.5, 1.0, 5.0, 10 mg/ml) of Xingding Injection. Ultraviolet radiation was administered to the cultured cells for 30 minutes. Westernblot assay was used to measure the expression of HSP 72 in the vascular endothelial cells. Results: There was no expression of HSP 72 in the cultured vascular endothelial cells without ultraviolet radiation, but there was some expression of HSP 72 after ultraviolet radiation. Xingding Injection of different concentrations could significantly improve the expression of HSP 72. The expression of HSP 72 in the vascular endothelial cells cultured in culture medium with 1.0 mg/ml Xingding Injection was the highest, and there was no more increase of expression when the concentration was higher, instead the expression decreased. Conclusion: Xingding Injection can protect the vascular endothelial cells from injury during stress. It may be one of its mechanisms in preventing and treating cardiocerebrovascular disorders.

  银杏素有“植物化石”之称,在治疗心脑血管病、内分泌疾病和妊娠病方面有良好的疗效。杏丁注射液是采用高科技手段从银杏叶中提取而成的制剂,具有明显的活血化瘀功效,对心脑血管疾病有较好的防治作用。研究表明[1],在紫外线、炎症、过氧化物、热休克、肿瘤坏死因子等作用下,受损细胞中的热休克蛋白(heat shock protein, HSP)基因转录活性升高和大量表达,HSP通过将变性的蛋白质折叠并介导其降解而发挥保护细胞的作用。为探讨杏丁注射液对血管内皮细胞的保护作用,作者观察了杏丁注射液对紫外线照射后猪主动脉血管内皮细胞HSP 72表达的影响,以期阐明其防治心脑血管疾病的机制。
  1  材料与方法
  1.1  实验材料
  1.1.1  药品与试剂  杏丁注射液,贵州益佰制药股份有限公司生产(批号:19990129)。胎牛血清、M199培养基,购自GibcoBRL公司;肝素和内皮细胞生长支持物,美国Sigma公司产品;丙烯酰胺/甲叉双丙烯酰胺、甘氨酸、三羟甲基氨基甲烷(Tris base)、十二烷基磺酸钠、四甲基乙二胺、二硫苏糖醇、苯甲基磺酰氟,Promega公司产品;兔抗猪HSP 72、辣根过氧化物酶(horseradish peroxidase, HRP)标记羊抗兔IgG和异硫氰酸荧光素标记羊抗兔IgG,北京中山生物公司产品;总蛋白质定量试剂盒,BioRad公司产品。其余均为国产试剂。
  1.1.2  仪器  YJ1450型医用净化工作台,哈尔滨净化设备公司;CO2孵箱,Forma公司;倒置显微镜、荧光显微镜(BX60型),Olympus公司;高速低温离心机,Sigma公司;Western blot垂直平板电泳槽、电泳仪,BioRad公司;恒温水浴箱,江苏太仓医疗仪器厂。
  1.2  实验方法
  1.2.1  猪主动脉血管内皮细胞原代培养及鉴定  选择正常屠宰的猪,用两把止血钳夹闭猪左锁骨上动脉和无名动脉,连同心脏和肺(带完整的胸膜)一同取出,迅速拿回实验室,紧贴主动脉弓切下主动脉段10~15 cm, DHanks液充分洗去血污,在动脉腔内注入0.25%胰蛋白酶,立即放入盛有300 ml磷酸盐缓冲液(phosphate buffer saline, PBS)的烧杯中,内含2%抗生素,37 ℃消化11~13 min。收集消化液并以M199培养液冲洗动脉管腔1~2遍,一并合入离心管,1 000 r/m
