Antioxidative activity of EPHPH on tissues and erythrocytes of rats
【Abstract】 AIM: To study the antioxidative activity of the alcoholic extract from Pseudostellaria heterophylla Pax et Hoffm (EPHPH) and its mechanism. METHODS: The peroxidation in homogenates of heart and liver and kidney of rats was induced by ・HO generation system Fe2++ascorbic acid. Malondialdehyde(MDA)was determined using TBA colorimetric method and superoxide(O2-)from zymosanstimulated neutrophils of rats was measured by NBT reduction method. H2O2caused hemolysis of erythrocytes was measured spectrometrically. RESULTS: MDA formation in homogenates of heart and liver and kidney of rats induced by ・HO generation system Fe2++ascorbic acid was inhibited differently by 6.7, 13.4, 26.8, 53.5, 107.0 and 214.0 μg/L EPHPH, whose IC50 that inhibited MDA formation in heart, liver and kidney was 57.0, 51.5 and 53.2 μg/L respectively, whose relation between quantity and efficiency showed all negative and whose relative coefficient was -0.891,-0.883 and -0.893 respectively (all P<0.001). O2- generation from zymosanstimulated neutrophils of rats was inhibited distinctly by EPHPH, whose IC50 that inhibited O2- generation was 83.1 μg/L, whose relation between quantity and efficiency showed negative and whose relative coefficient was -0.894(P<0.001). H2O2caused hemolysis of erythrocytes was inhibited distinctly by EPHPH whose IC50 that inhibited hemolysis of erythrocytes was 86.5 μg/L, whose relation between quantity and efficiency showed negative and whose relative coefficient was -0.901 (P<0.001). CONCLUSION: EPHPH exerts its effect on antioxidation by scavenging ・HO, O2- and H2O2 in the biological system.

【摘要】 目的: 研究太子参醇提物的抗氧化活性及其机制. 方法: 用・HO生成系统Fe2++抗坏血酸诱导大鼠心、肝、肾组织匀浆脂质过氧化,TBA比色法测定MDA含量;NBT还原法测酵母多糖A刺激大鼠中性粒细胞产生的O2-;分光光度法测H2O2诱发的大鼠红细胞溶血度. 结果: 6.7, 13.4, 26.8, 53.5, 107.0和214.0 μg/L 太子参醇提物不同程度抑制Fe2++抗坏血酸诱导的大鼠心、肝、肾MDA生成,其抑制心、肝、肾MDA生成的IC50分别为57.0, 51.5及53.2 μg/L, 其量效关系均呈负相关,相关系数分别为-0.891, -0.883, -0.893 (P均<0.001);不同程度抑制酵母多糖A刺激大鼠中性粒细胞生成O2-及红细胞氧化溶血,IC50分别为83.1和86.5 μg/L,其量效关系均呈负相关,相关系数分别为-0.894和-0.901 (P均<0.001). 结论: 太子参醇提物通过清除・HO, O2-及H2O2而发挥抗氧化活性.

0引言
太子参(Pseudostellaria heterophylla Pax et Hoffm, PHPH)为传统中药,有补肺健脾功效. 近年有研究表明太子参总提物对环磷酰胺所致小鼠T、B淋巴细胞转化功能低下及白细胞吞噬功能降低有显著对抗作用,并能增加外周血白细胞数,其水提物有抗氧化作用[1]. 我们通过实验将太子参醇提物(EPHPH)对大鼠组织和红细胞的抗氧化活性及其机制进行了探讨.
1材料和方法
1.1材料
EPHPH用50 mL/L DMSO溶解,硫代巴比妥酸(TBA)为上海试剂二厂产品,氯化硝基四氮唑蓝(NBT)为上海前进试剂厂产品,酵母多糖A为Sigma产品,其它试剂均为进口或国产分析纯. Wistar大鼠40只,雌雄不拘,体质量(210±20) g,由兰州医学院实验动物中心提供. 药材购自本院药房.
1.2方法
1.2.1EPHPH的制备用回流提取法,将药材适度粉碎,800 mL/L乙醇浸泡24 h,装入瓶内,约为容量的1/2,溶剂浸过药材表面约2 cm,水浴中加热回流,放冷过滤,第1次回流1 h,第2及第3次各约30 min,回收乙醇,60℃干燥即得. 样品主 1 2 3 下一页
